Effect of Extraction Methods on Physicochemical Characteristics and Antioxidant Activities of Crude

Abstract
This study was conducted to examine the effect of extraction methods on physicochemical characteristics and antioxidant activities of crude polysaccharides isolated from Ecklonia cava. The yield of extract ranged from 6.67 to 20.47%. The total polyphenol and total flavonoid contents of viscozyme extract were 16.56 g/100 g, 13.25 g/100 g, respectively. The total sugar content of viscozyme extract was 74.16 g/100 g, and the total protein content of neutrase extract was 20.95 g/100 g. The DPPH and ABTS radical scavenging activities of viscozyme extract at 1,000 ¥ìg/mL were 46.03% and 43.16%. The ferric-reducing antioxidant power of viscozyme extract at 1,000 ¥ìg/mL was 1.21 mM. The functionalities of extracts were investigated with L-132 cell lines. The crude polysaccharides not showed the toxicity on L-132 cell line in 100~1,000 ¥ìg/mL. The viscozyme extract (1,000 ¥ìg/mL) performed better than other extracts protective effects against oxidative stess in L-132 cells (86.47%) which was more potent than other extracts. There results suggest that the viscozyme extract may be a useful functional food material in the food industry.
